et al., 1952). In liver disease or folic acid deficiency, abnormal quantities ofthe metabolites, urocanic acid and formiminoglutamic acid (Figlu), appear in the urine following an oral dose of the amino acid L-histidine (Luhby, Cooperman, and Teller, 1959; Spray and Witts, 1959; Kohn, Mollin, and Rosenbach, 1961; Carter, Schaffner, and Heller, 1960; Bennett and Chanarin, 1962; Merritt et al., 1962). In mammalian liver, the major pathway of histidine metabolism is its degradative conversion via urocanic acid to glutamic acid (Fig. 1). The abnormal excretion of Figlu in heart failure (Grasbeck, Bjorksten, and Nyberg, 1961) was confirmed by one of us (Rose, 1964), and in addition excess urocanic acid was noted. It therefore appeared that the hepatic congestion of heart failure resulted in defective histidine metabolism, and it was decided to study this in more detail. We have therefore estimated the excretion of urocanic acid following an oral dose of histidine in patients with congestive cardiac failure due to

Urinary excretion of histidine derivatives in megaloblastic anaemia and other conditions and comparison with the folic acid clearance test.

Urinary excretion of urocanic acid and formimino-glutamic acid in abnormal amounts following oral doses of histidine has been observed not only in most cases of megaloblastic anaemia, but in iron-deficiency anaemia, haemolytic anaemia, and neoplastic disease. The urinary excretion of these substances is increased in those disease states in which an abnormally rapid rate of removal from the plas...

Relative importance of formiminoglutamic and urocanic acid excretion after a histidine load.

A thin-layer chromatographic method for the simultaneous assessment of formimino-glutamic (Figlu) and urocanic acid excretion after loading with 15 g. histidine has been applied to 15 control subjects and to 49 selected patients. Upper limits of normal excretion were determined for urocanic acid and Figlu alone (15 mg. and 30 mg/8 hr. respectively), and for the combined metabolites (40 mg./8 hr...

Urinary urocanic acid in man: the identification of urocanic acid and the comparative excretions of urocanic acid and N-formiminoglutamic acid after oral histidine in patients with liver disease.

The amino acid L-histidine is obtained by man primarily from dietary protein. The essential need for this amino acid, however, has not been demonstrated conclusively (1). Previous studies have indicated that the major pathways of utilization of L-histidine are by way of incorporation into body protein, excretion into the urine, and metabolism to glutamic acid. It is believed that the pertinent ...

Detection by paper chromatography of imidazoles, including hydantoin-5-propionic acid, in urine after histidine dosage.

Simple paper chromatography of urine obtained in the Figlu test has been found satisfactory for determining excess excretion of the imidazole metabolites of histidine. It has been confirmed that examination for these in addition to formiminoglutamic acid is necessary for a full assessment of this test. When excretion of the latter is not abnormal in folic-acid and/or vitamin B(12) deficiency, t...

Histidinaemia: a child and his family.

Histidinaemia, an inherited disorder of histidine metabolism, was first described by Ghadimi, Partington, and Hunter in 1961. The principal metabolic pathway for histidine is through urocanic acid and formiminoglutamic acid (FIGLU) to glutamic acid (Fig. 1). Zannoni and La Du (1963) demonstrated that histidase, the enzyme necessary for the conversion of histidine to urocanic acid, was absent fr...
